چکیده

These Comparative genomics and genomic tools have been used to identify virulence factors and genes involved in environmental persistence of pathogens. However, a major stumbling block in the genomics revolution has been the large number of genes with unknown function that have been identified in every organism sequenced to date. Over 1740 bacterial genome sequences are currently available in public databases and over 5230 are in progress, representing hundreds of species as well as multiple strains of the same species. The study of these genomes by both computational and experimental approaches has significantly advanced our understanding of the physiology and pathogenicity of many microbes and provided insights into the mechanisms and history of genome evolution. Several ‘‘postgenomic’’ methods have been utilized to identify genes that are essential for bacterial growth or pathogenesis. Here we demonstrate the utility of several DNA and protein sequence comparison tools to interpret the information obtained from several genome projects. Comparisons are presented between closely related strains of Staphylococcus aureus and S. epidermidis spp. The comparative genome analysis will generate a wealth of data to compare pathogenic strains with varying levels of pathogenicity, which in turn may reveal mechanisms by which the pathogen may adapt to a particular host.

Comparative genomics of human stem cell factor (SCF)

Stem cell factor (SCF) is a critical protein with key roles in the cell such as hematopoiesis, gametogenesis and melanogenesis. In the present study a comparative analysis on nucleotide sequences of SCF was performed in Humanoids using bioinformatics tools including NCBI-BLAST, MEGA6, and JBrowse. Our analysis of nucleotide sequences to find closely evolved organisms with high similarity by NCB...

Identification of Gene Clusters Associated with Host Adaptation and Antibiotic Resistance in Chinese Staphylococcus aureus Isolates by Microarray-Based Comparative Genomics

A comparative genomic microarray comprising 2,457 genes from two whole genomes of S. aureus was employed for the comparative genome hybridization analysis of 50 strains of divergent clonal lineages, including methicillin-resistant S. aureus (MRSA), methicillin-susceptible S. aureus (MSSA), and swine strains in China. Large-scale validation was confirmed via polymerase chain reaction in 160 repr...

Comparative Genomic Assessment of Novel Broad-Spectrum Targets for Antibacterial Drugs

Single and multiple resistance to antibacterial drugs currently in use is spreading, since they act against only a very small number of molecular targets; finding novel targets for anti-infectives is therefore of great importance. All protein sequences from three pathogens (Staphylococcus aureus, Mycobacterium tuberculosis and Escherichia coli O157:H7 EDL993) were assessed via comparative genom...

Factors characterizing Staphylococcus epidermidis invasiveness determined by comparative genomics.

Virulence mechanisms of the leading nosocomial pathogen Staphylococcus epidermidis are poorly understood. We used microarray-based genome-wide comparison of clinical and commensal S. epidermidis strains to identify putative virulence determinants. Our study revealed high genetic variability of the S. epidermidis genome, new markers for invasiveness of S. epidermidis, and potential targets for d...

Comparative genomics of Staphylococcus aureus musculoskeletal isolates.

Much of the research aimed at defining the pathogenesis of Staphylococcus aureus has been done with a limited number of strains, most notably the 8325-4 derivative RN6390. Several lines of evidence indicate that this strain is unique by comparison to clinical isolates of S. aureus. Based on this, we have focused our efforts on two clinical isolates (UAMS-1 and UAMS-601), both of which are hyper...
